Establishing a platform for generation of E.coli based protein expression systems

Project Description

The goal of this Master’s thesis is to design and validate a structured platform for screening and establishing of E. coli-based protein expression systems protein therapeutics. The project incorporates theoretical analysis and planning followed by hands on lab work to demonstrate the capability of the platform

A preliminary project layout could be:

1.       Theoretical Evaluation

-          Comparative analysis of E. coli strains, expression vectors, and culture formats

-          Selection of suitable analytical methods for expression analysis

-          Literature-based assessment of key parameters affecting yield and quality

2.       Proof-of-Concept Laboratory Work

-          Application of the designed workflow using selected model proteins

-          Hands-on work from DNA sequence to expression-ready E. coli cell lines

-          Evaluation of expression yield and protein quality

The ideal outcome is an effective platform that starts with a theoretical protein or DNA sequence and results in a transformed E. coli strain capable of producing the target protein at yields and quality levels suitable for manufacturing. During the thesis work you will gain practical experience in molecular biology methods, microbial bioprocessing in different scales, protein isolation and analysis. The project is located at NorthX Innovation laboratory in Matfors and is expected to have approximately 20 weeks’ duration and the start date is flexible from the beginning of September.

Who Are We Looking For?

We are looking for a highly motivated and self-driven Master student who:

-          Has a theoretical background in biotechnology, molecular biology, and protein expression

-          Excels in finding, analyzing, and applying scientific information

-          Has a keen interest in molecular biology and microbiology techniques

-          Previous experience with E. coli expression systems, recombinant protein production, or analytical methods (e.g., SDS-PAGE, Western blot) is a plus but not mandatory.

-          Holds a positive attitude and a curious mind
